Misinterpreting My Roommate's Tone

So I live with my two best friends, I've known them for years, and we moved in together last year. One of my friends is super upbeat and straightforward with stuff but has a lot of medical stuff going on so he isn't always as chipper as he would be otherwise. I have a bad time understanding some people's tone. I end up reading too much into it or misinterpreting it. And half the time I end up thinking that the tone is because of me. That they're mad or something. My roommate, he has a specific tone that he gets when he isn't feeling well, and it sounds a lot like he's annoyed or talking down to me and I know that he isn't and he just probably isn't feeling well but I may be wrong? And i ask if everything is fine he has gotten to the point where he's like "look it really feels like I'm walking on eggshells with you all the time. Like anything I saw will set you off" Its really stressing me out and I don't know what to do.

Be honest with them, they’re your best friend. I’d say that same thing you said here. Look best friend, let’s talk this out I feel xyz how do you feel, what can we do to work it out. Honestly is the best policy 💕
